SUPT7L encodes subunit gamma of the STAGA (SPT3-TAF9-GCN5 Acetyltransferase) transcriptional coactivator complex, a multi-protein assembly involved in regulation of gene expression via chromatin modification and histone acetylation[2][3][5]. SUPT7L serves as a structural scaffold in the STAGA complex, critical for its assembly and function, and participates in transcriptional coactivation, including transcription factor binding, acetyltransferase activity, and recruitment of the transcription machinery[1][2][3][5]. Loss-of-function mutations in SUPT7L are linked to rare genetic syndromes such as generalized lipodystrophy with progeroid features, implicating it in genome stability and developmental pathways[4]. Dysregulation or aberrant expression of STAGA subunits (including SUPT7L and related components) has associations with cancer, particularly adenocarcinoma, through effects on transcriptional repression/activation and chromatin remodeling[3][4]. There are no known approved drugs that directly target SUPT7L, and it is not currently considered a canonical therapeutic target such as a receptor, enzyme, or transporter[3][4].
