SSTR5 antisense RNA 1 (SSTR5-AS1) is a long non-coding RNA and natural antisense transcript located at the SSTR5 locus on chromosome 16[3]. SSTR5-AS1 regulates the expression of the neighboring SSTR5 gene, which encodes the somatostatin receptor subtype 5, through mechanisms involving DNA methylation and chromatin modification[1][3]. In neuroendocrine tumors, SSTR5-AS1 and SSTR5 levels are tightly correlated, and SSTR5-AS1 silencing reduces SSTR5 expression and can increase cellular proliferation and tumor aggressiveness, as well as alter the response to the somatostatin analogue pasireotide[1]. In laryngeal and esophageal carcinomas, SSTR5-AS1 acts as a tumor suppressor by inhibiting proliferation, migration, and invasion, potentially through interactions with chromatin-modifying proteins like MLL3 and TET1[2][3]. Thus, SSTR5-AS1 plays an important epigenetic regulatory role but is not considered a canonical therapeutic target like receptors or enzymes; its clinical relevance is more as a biomarker and regulatory RNA influencing targetable pathways.
