The ST3GAL4 mRNA 3' untranslated region (3'UTR) is a critical regulatory segment of the messenger RNA encoding the ST3 beta-galactoside alpha-2,3-sialyltransferase 4 enzyme. This region contains binding sites for microRNAs (miRNAs) and RNA-binding proteins that modulate the stability and translation efficiency of the ST3GAL4 transcript. In various cancers, including gastric and pancreatic cancer, the dysregulation of this UTR-mediated control leads to the overexpression of ST3GAL4, which in turn increases the synthesis of sialyl-Lewis X (sLeX) epitopes on the cell surface. These epitopes are essential for selectin-mediated adhesion and subsequent metastatic spread. Consequently, the ST3GAL4 mRNA 3'UTR is an emerging therapeutic target for RNA-based interventions, such as miRNA mimics (e.g., miR-370) or antisense oligonucleotides, aimed at reducing enzyme levels to inhibit tumor progression and metastasis. Beyond oncology, the regulation of this region is also implicated in inflammatory conditions like osteoarthritis and the synthesis of receptors for avian influenza viruses [PubMed: 33080315, NIH]. Therapeutic strategies targeting this UTR aim to restore normal sialylation patterns and prevent pathological cell-cell interactions. Challenges in targeting this region include ensuring specificity to avoid off-target RNA interactions and achieving effective delivery to target tissues.
