Overview

Stem cell antigen-1 (Sca-1), primarily known as Lymphocyte antigen 6A (Ly6A), is a glycosylphosphatidylinositol (GPI)-anchored cell surface protein belonging to the Ly6 family. It is a critical marker in murine research, used extensively to identify and enrich hematopoietic stem cells (HSCs) and other tissue-resident progenitors. Beyond its function as a biomarker, Sca-1 regulates biological processes such as cell self-renewal, proliferation, and differentiation by participating in lipid raft-associated signaling complexes. In pathological contexts, Sca-1/LY6A is often overexpressed in various cancers, including mammary and pituitary tumors, where it promotes tumor migration and stemness. While traditionally associated with mice, the discovery of a human candidate ortholog (LY6A) has increased its relevance as a potential therapeutic target in human oncology. Drugs like histone deacetylase inhibitors (HDACis) and interferons are known to modulate its expression, offering avenues for manipulating stem cell outcomes or immune responses.
