Overview

HPN-102 target mRNA refers to the messenger RNA of the Sterol Regulatory Element-Binding Transcription Factor 1 (SREBF1), which is the primary target of the investigational GalNAc-conjugated siRNA drug HPN-102 developed by Hepanova. SREBF1 is a critical transcription factor that regulates the expression of genes involved in cholesterol and fatty acid biosynthesis, playing a central role in hepatic lipogenesis. In the context of metabolic dysfunction-associated steatohepatitis (MASH), overactivation of the SREBF1 pathway leads to excessive lipid accumulation (steatosis), inflammation, and subsequent liver fibrosis. By utilizing RNA interference (RNAi), HPN-102 specifically targets and degrades SREBF1 mRNA in hepatocytes, thereby reducing the production of the SREBP-1 protein and downregulating the lipogenic program. This therapeutic approach aims to alleviate liver fat accumulation and arrest the progression of fibrosis in patients with MASH.
