Streptococcus pneumoniae capsular polysaccharides are the primary virulence factors of the pneumococcus bacterium, forming a protective outer layer that inhibits phagocytosis by host immune cells [4, 10]. These polysaccharides are chemically diverse, with over 100 distinct serotypes identified to date, each eliciting a specific immune response [4, 10]. In the GSK2830930A (also known as ASP3772) formulation, 24 specific capsular polysaccharides are included: serotypes 1, 2, 3, 4, 5, 6A, 6B, 7F, 8, 9N, 9V, 10A, 11A, 12F, 14, 15B, 17F, 18C, 19A, 19F, 20, 22F, 23F, and 33F [1, 2]. Unlike traditional conjugate vaccines, GSK2830930A utilizes the Multiple Antigen Presenting System (MAPS) technology, which non-covalently links the polysaccharides to protein antigens to stimulate both humoral and cellular immunity [1, 3]. The target's role in disease involves facilitating bacterial colonization and invasion of the bloodstream or central nervous system, leading to conditions such as pneumonia, sepsis, and meningitis [3, 10]. By targeting these polysaccharides, the vaccine induces opsonophagocytic antibodies that facilitate the clearance of the bacteria by the host immune system [1, 3]. This broad-spectrum approach aims to provide protection against a significant majority of serotypes responsible for invasive pneumococcal disease globally [1, 2]. The inclusion of additional serotypes like 2, 9N, 17F, and 20 expands coverage beyond existing 20-valent vaccines [2, 6]. Clinical evaluation of these targets focuses on the induction of functional antibodies measured by opsonophagocytic activity assays [1, 3]. Overall, these polysaccharides represent critical targets for preventing the high morbidity and mortality associated with pneumococcal infections [3, 10].
