Streptococcus pneumoniae polysaccharide antigens are complex carbohydrate structures that form the outer capsule of the bacterium *S. pneumoniae*. These capsular polysaccharides are highly variable, with over 95 distinct serotypes identified. The capsule is the primary virulence factor for *S. pneumoniae*, enabling it to evade host immune responses. To enhance immunogenicity, these polysaccharides are chemically conjugated to carrier proteins such as CRM197—a non-toxic mutant of diphtheria toxin—creating glycoconjugate vaccines that induce robust T-cell-dependent immune responses even in infants. When used in vaccines purified capsular polysaccharides from selected clinically relevant *S. pneumoniae* serotypes are covalently linked to CRM197. Upon administration, these conjugates stimulate B cells via both T-cell-dependent and independent pathways, leading to high-affinity antibody production and immunological memory formation.
