Streptolysin O (SLO) is a potent, oxygen-labile, cholesterol-dependent cytolysin (CDC) secreted by Streptococcus pyogenes, also known as Group A Streptococcus (GAS). It functions by binding to cholesterol in host cell membranes, where it oligomerizes to form large transmembrane pores (approximately 25-30 nm in diameter), leading to cell lysis and death (Source: UniProt P0C0I4). Beyond direct cytotoxicity, SLO facilitates the translocation of other bacterial virulence factors, such as NAD-glycohydrolase, into the host cytosol to subvert immune responses and promote intracellular survival (Source: PubMed 11544344). In clinical practice, SLO is a critical diagnostic marker; elevated anti-streptolysin O (ASO) titers indicate recent GAS infection and are used to diagnose post-streptococcal complications like acute rheumatic fever and post-streptococcal glomerulonephritis (Source: StatPearls NBK553089). While not a traditional human receptor, SLO is a target for therapeutic intervention using intravenous immunoglobulin (IVIG) to neutralize toxins in severe, life-threatening infections like streptococcal toxic shock syndrome (Source: PubMed 15106084). Current research also explores SLO as a potential vaccine component and a target for neutralizing monoclonal antibodies or small-molecule inhibitors to mitigate tissue damage during invasive GAS infections (Source: PubMed 28416514).
