Overview

Superoxide dismutase [Mn], mitochondrial (SOD2) is a nuclear-encoded enzyme essential for maintaining the balance of reactive oxygen and nitrogen species within the mitochondria (UniProt P04179). It catalyzes the conversion of superoxide radicals, which are toxic byproducts of oxidative phosphorylation, into hydrogen peroxide and molecular oxygen, thereby preventing oxidative damage to cellular components (PubMed: 29453904). This Mn-mediated redox chemistry is a critical defense mechanism against oxidative stress-induced cell death and inflammation. In clinical settings, manganese-based SOD mimetics like avasopasem manganese are used to replicate this activity, particularly to protect healthy tissues from the damaging effects of superoxide bursts during radiation therapy (PubMed: 32813454). These drugs utilize a stable manganese coordination complex to facilitate the catalytic removal of superoxide, offering a therapeutic approach to diseases characterized by excessive ROS production, such as cancer and inflammatory disorders.
