SNAP-25 is a key component of the SNARE complex, which is essential for mediating vesicle fusion with target membranes during exocytosis, particularly neurotransmitter release at synapses. It interacts with synaptobrevin (VAMP2) and syntaxin (STX1A) to form a tight complex that drives membrane fusion. SNAP-25 also regulates synaptic vesicle docking, priming, and triggering. Furthermore, it modulates voltage-gated calcium channels and is involved in postsynaptic processes such as dendritic spine morphogenesis. Dysfunction of SNAP-25 is linked to several neurodevelopmental and neuropsychiatric disorders.
Currently, there are no drugs that directly target SNAP-25 for therapeutic intervention. However, botulinum toxins indirectly target SNAP-25 by cleaving it and preventing its function.
