Overview

The SSX2-derived peptide–MHC class I complex is a specialized immunological target consisting of a peptide fragment from the Synovial Sarcoma, X Breakpoint 2 (SSX2) protein bound to a Major Histocompatibility Complex (MHC) class I molecule, typically HLA-A*02:01 (UniProt: O60225; PubMed: 12615711). SSX2 is a cancer-testis antigen (CTA) that is normally expressed only in the testis but becomes aberrantly re-expressed in various cancers, including synovial sarcoma, melanoma, and hepatocellular carcinoma (PubMed: 34123841). Because the testis is an immunologically privileged site lacking MHC class I expression, the presentation of SSX2 peptides on tumor cells creates a highly tumor-specific target for the immune system (PubMed: 12615711; ClinicalTrials.gov: NCT04917523). Therapeutic strategies targeting this complex primarily involve T-cell receptor (TCR) engineered T-cell therapies, which are designed to recognize the specific peptide-MHC configuration and induce targeted cell lysis (ClinicalTrials.gov: NCT04917523). These therapies aim to exploit the restricted expression pattern of SSX2 to minimize off-target effects on healthy somatic tissues while providing a potent anti-tumor response (PubMed: 34123841).
