SSX4B (Synovial sarcoma, X breakpoint 4B) is a protein encoded by the *SSX4B* gene, a member of the SSX cancer/testis antigen family. SSX4B is nearly identical to its paralog SSX4, produced by a segmental duplication on the X chromosome, and is thought to function as a transcriptional repressor or modulatory protein in the nucleus[2][6][7]. Expression of SSX4B (and closely related SSX proteins) is normally restricted to germline tissues but is reactivated in various cancers, notably synovial sarcoma, melanoma, and other solid tumors[2][4][6][8]. Functional studies show SSX4B supports tumor cell proliferation, survival, migration, and invasion, often by influencing pathways such as MAPK/Erk and Wnt[8]. SSX4B is a potent tumor-associated antigen, capable of eliciting spontaneous immune responses in cancer patients; thus, it is regarded as a promising target for cancer vaccines and adoptive T cell therapies[2][4][8]. No direct small-molecule inhibitors or approved therapies currently target SSX4B, but it is a validated immunotherapeutic target with ongoing research interest[2][6][8].
Immunotherapeutic strategies: Cancer vaccine or T cell-based immunotherapy, where the mechanism is to elicit immune responses (especially CD4+ and cytotoxic T lymphocyte responses) to tumor cells expressing SSX4B. Putative inhibition: Knockdown of SSX4B (and SSX4) by RNA interference results in reduced tumor cell proliferation and survival (mechanistically through modulation of MAPK/Erk and Wnt signaling pathways).
