Overview

Syntabulin (SYBU) is a microtubule-associated, peripheral membrane adaptor protein involved in the intracellular transport of key vesicular cargoes, including syntaxin-1-containing vesicles and mitochondria, especially within neurons[1][2][3][6][7]. Syntabulin binds to both kinesin motor proteins and syntaxin vesicles, serving as a crucial "linker" that attaches cargoes to microtubules, thereby enabling their anterograde transport along axons[3][7]. The protein also contains functionally specialized domains: a kinesin-binding domain (N-terminus), a central syntaxin-binding domain, and a C-terminal mitochondrial-binding domain, underscoring its multifaceted role in axonal transport and presynaptic function[4][6]. Syntabulin's functions are critical to neuronal health and synapse formation, and the protein is expressed in a range of neuronal and non-neuronal tissues, reflecting its diverse cellular functions[2][5][6]. Note: Syntabulin is a trafficking scaffold protein, not a classic therapeutic target (receptor, enzyme, ion channel, etc.), and no clinically approved drugs directly target it. Its biological relevance is mainly in synaptic biology and neurological disease research[2][6].
