Overview

The synthetic super-enhancer-associated transcription factor complex is a multimeric assembly of proteins, primarily featuring SOX2 and SOX9, that governs the identity and survival of glioblastoma stem-like cells (GSCs) (Koeber et al., 2026). These transcription factors act as master regulators by binding to large clusters of regulatory DNA known as super-enhancers, which are disproportionately active in aggressive cancer cells compared to healthy tissue (Garros-Regulez et al., 2016). In therapeutic applications, synthetic super-enhancers (SSEs) are engineered to serve as artificial docking sites for this complex, enabling the selective expression of therapeutic genes only within GSCs (Trogenix, 2024). This approach, exemplified by the Odysseus platform and the candidate TGX-007, allows for the targeted delivery of cytotoxic or immunomodulatory payloads while minimizing effects on healthy brain tissue (ed.ac.uk, 2024). By exploiting the unique transcriptional addiction of glioblastoma cells, this target offers a path toward high-precision gene therapy for aggressive brain tumors (allsci.com, 2026).
