T-box transcription factor 3 (TBX3) is a member of the evolutionary conserved T-box family of transcription factors, playing a fundamental role in the embryonic development of organs such as the heart, mammary glands, and limbs [2, 3]. While its expression is strictly regulated and minimal in normal adult tissues, TBX3 is frequently upregulated in a wide array of malignancies, including breast cancer, melanoma, and various sarcomas [1, 4, 16]. In these contexts, it functions as a potent oncoprotein by transcriptionally repressing key tumor suppressors like p14ARF and p21, thereby enabling cancer cells to bypass senescence and avoid apoptosis [1, 5, 9, 13]. Beyond cancer, TBX3 is implicated in developmental disorders like Ulnar-Mammary Syndrome and adult conditions such as rheumatoid arthritis and obesity [2, 4, 18]. Therapeutic strategies targeting TBX3 are currently being explored, with a focus on small molecules like niclosamide and pyrvinium pamoate that can downregulate its protein levels or disrupt its essential co-factor interactions [11, 15]. As direct inhibition of transcription factors is pharmacologically challenging, these indirect approaches represent the primary avenue for clinical development [1, 7, 9].
