T cell factor 1 (TCF-1), encoded by the Tcf7 gene, is a T cell-specific DNA-binding transcription factor that acts as a central regulator of T cell development, differentiation, and function across multiple stages and lineages, including thymocyte maturation from early thymic progenitors to CD4+ and CD8+ T cells, and regulation of effector, memory, follicular helper, and regulatory T cell subsets. It promotes T cell identity by organizing three-dimensional genome structure, maintaining chromatin accessibility at direct binding sites, and cooperating with factors like Lef1, β-catenin, HEB, Runx1, Ikaros, and Foxp3 to activate T lineage genes while repressing non-T lineage genes; it also possesses intrinsic histone deacetylase activity to reduce chromatin accessibility and suppress certain gene expression. TCF-1 functions downstream of Notch and Wnt/β-catenin signaling but has significant Wnt-independent roles, such as preserving stemness in exhausted CD8+ T cells during chronic infection or cancer. Loss of TCF-1 blocks early T cell specification, impairs lineage choices, and alters responses to infection, autoimmunity, and tumors.
