T-cell protein tyrosine phosphatase (TC-PTP), encoded by the PTPN2 gene, is a non-receptor protein tyrosine phosphatase that acts as a critical negative regulator of various cytokine signaling pathways, most notably the JAK-STAT axis. By dephosphorylating key signaling molecules such as JAK1, JAK3, STAT1, STAT3, and STAT5, TC-PTP modulates the cellular response to inflammatory stimuli and growth factors. Research highlighted by Jan Cools and colleagues in the journal Blood (2011) established PTPN2 as a major tumor suppressor in T-cell acute lymphoblastic leukemia (T-ALL), where its loss leads to hyperactivated kinase signaling and enhanced leukemogenesis. Beyond leukemia, TC-PTP has emerged as a significant target in cancer immunotherapy, as its inhibition or degradation can sensitize tumor cells to interferon-gamma and bolster the anti-tumor activity of T-cells. Consequently, the development of small-molecule inhibitors and PROTAC degraders targeting TC-PTP represents a novel therapeutic approach to treating solid tumors and hematological malignancies by reversing the suppression of the immune microenvironment.
