Overview

The T-cell receptor (TCR) on T lymphocytes recognizing EO2318-derived peptide–MHC complexes is a specific immune receptor that mediates the recognition of microbiome-derived antigens mimicking tumor proteins. EO2318 is a synthetic peptide mimic (OncoMimic) derived from commensal gut bacteria that shares high structural homology with a specific epitope of the human tumor-associated antigen FOXM1 (Forkhead box M1) (Magalhaes et al., 2024). This TCR is primarily expressed on CD8+ cytotoxic T cells and recognizes the EO2318 peptide when it is presented by the Major Histocompatibility Complex (MHC) class I molecule HLA-A*02:01 (Enterome, 2025). In therapeutic applications, vaccines such as EO2401 and EO4010 are used to deliver the EO2318 peptide, thereby activating and expanding a pool of memory T cells that were previously primed by the gut microbiome (BioRxiv, 2024). Because of the molecular mimicry between the bacterial peptide and the tumor antigen, these expanded T cells can cross-react with FOXM1-expressing cancer cells, such as those found in glioblastoma and colorectal cancer (NIH, 2025). This strategy is designed to overcome the immune tolerance typically associated with self-antigens, providing a potent and durable anti-tumor response (ClinicalTrials.gov, 2025).
