Overview

The T-cell receptor beta variable region (TCR Vβ) is a highly polymorphic domain of the T-cell receptor responsible for the recognition of peptide antigens presented by major histocompatibility complex (MHC) molecules (StatPearls, 2023). This region is formed by the somatic recombination of variable (V), diversity (D), and joining (J) gene segments, providing the structural diversity necessary for the adaptive immune system to encounter a vast array of pathogens (PubMed, PMID: 10880530). Beyond its role in normal immunity, the Vβ region is the primary binding site for superantigens, which cross-link the TCR with MHC class II molecules, leading to non-specific, massive T-cell activation and potentially fatal cytokine storms (NIH, 2022). In therapeutic development, the Vβ region is targeted to treat T-cell malignancies through the use of Vβ-specific monoclonal antibodies or CAR-T cells that selectively eliminate clonal populations of cancerous T cells while sparing the rest of the immune repertoire (Nature Communications, 2017). Monitoring the Vβ repertoire also serves as a vital biomarker for assessing immune recovery after bone marrow transplantation or response to immunotherapy (UniProt, 2024).
