Overview

The T-cell receptor delta chain V delta 2 (Vδ2-TCR) is a component of the γδ T-cell receptor complex that is prominently expressed on a major subset of human peripheral blood γδ T cells, particularly those bearing a Vγ9 TCR chain (Vγ9Vδ2 T cells)[3][4]. The Vδ2-TCR enables γδ T cells to recognize a range of non-peptide phosphoantigen molecules, which accumulate during microbial infection or cellular transformation (malignancy)—this process is mediated in concert with the butyrophilin 3A1/BTN2A1 complex[5][6][7]. Upon activation, Vδ2 TCR+ T cells rapidly exert cytotoxic, cytokine-secreting, and antigen-presenting functions, contributing to antimicrobial defense and antitumor immunity[3][4][7][8]. Vδ2 TCRs show both innate-like and adaptive features, depending on Vγ pairing and antigen exposure[3][4]. Their unique biology and reactivity make the Vδ2-TCR an attractive, but complex, target for immunotherapy and biomarkers in cancer and infectious disease settings[1][5][7].
