Overview

The T cell receptor-Major Histocompatibility Complex (TCR-MHC) recognition machinery is the fundamental unit of the adaptive immune system responsible for identifying foreign antigens. This complex consists of the TCR (typically an alpha-beta heterodimer) associated with the CD3 signaling apparatus, which recognizes short peptides presented by MHC (or HLA in humans) molecules on the surface of cells (Janeway's Immunobiology, 9th Ed). This interaction is highly specific and determines the activation, proliferation, and effector functions of T cells. In oncology, this machinery is targeted by TCR-engineered T cells (TCR-T) and bispecific antibodies to direct immune responses against tumor-specific antigens (Nature Reviews Drug Discovery, 2019). Conversely, in autoimmunity and transplantation, drugs target this machinery to suppress unwanted T-cell activation and prevent tissue destruction (New England Journal of Medicine, 2020). The structural integrity and binding affinity of the TCR-pMHC interface are critical factors in determining the efficacy and safety of immunotherapies.
