Overview

The T-cell receptor (TCR) on CD4+ T-cells is a highly diverse, heterodimeric protein complex (usually αβ chains) expressed on the surface of CD4+ T-lymphocytes, also known as helper T-cells. TCRs recognize antigens presented as peptides bound to major histocompatibility complex (MHC) class II molecules on antigen-presenting cells. Each TCR is generated through V(D)J recombination, endowing it with unique specificity toward foreign and self-peptides. Engagement of the TCR with antigen/MHC triggers intracellular signaling cascades, primarily through association with the CD3 complex (containing ITAMs), ultimately leading to T-cell activation, differentiation into effector or memory cells, and cytokine production necessary for orchestrating immune responses. CD4 serves as a coreceptor helping stabilize MHC II interaction and recruiting Lck kinase for signal transduction. Dysregulation or aberrant activation of TCRs on CD4+ T-cells can drive autoimmune diseases, contribute to pathogenic inflammation, or be harnessed for immunotherapy. The molecular pathways and specificity of TCR action in CD4+ T cells make it a critical node for therapeutic interventions in immunology, autoimmunity, infection, and oncology.
