Overview

The T-cell receptor (TCR) recognizing RCC-26 tumor-associated antigen peptides in the HLA-A*0201 context is a specialized immune receptor used in the development of adoptive T-cell therapies for renal cell carcinoma (RCC). It was originally isolated from tumor-infiltrating lymphocytes (TILs) of a patient (patient 26) who exhibited a significant clinical response to the RCC-26 tumor cell line (Schendel et al., 1993). This TCR specifically recognizes peptides derived from tumor-associated antigens (TAAs) or neoantigens that are overexpressed in RCC cells and presented by the HLA-A*0201 major histocompatibility complex (MHC) molecule (Leisegang et al., 2010). In therapeutic applications, T cells are genetically engineered to express this TCR, enabling them to target and destroy RCC cells with high specificity. While early versions of this TCR were patient-specific, research has focused on identifying shared antigens that allow for broader clinical utility. The use of TCR-RCC26 in therapy requires careful screening for HLA-A*0201 positivity and monitoring for potential off-target effects or cross-reactivity with healthy tissues (Pohla et al., 2000). This target represents a key component in the precision medicine approach to treating solid tumors through TCR-engineered T-cell (TCR-T) therapy.
