Overview

T-cell receptors (TCRs) specific for LY6K, VEGFR1, and VEGFR2 epitopes presented by HLA-A*2402 are immune recognition molecules designed to target a combination of tumor-associated antigens (TAAs) and angiogenesis-related factors. LY6K (Lymphocyte antigen 6 complex, locus K), also known as URLC10, is a cancer-testis antigen frequently overexpressed in various solid tumors, including esophageal, lung, and gastric cancers, where it promotes cell proliferation and survival (PubMed: 25340007). VEGFR1 and VEGFR2 (Vascular Endothelial Growth Factor Receptors 1 and 2) are critical mediators of tumor angiogenesis expressed on vascular endothelial cells (PubMed: 25340007, NCI Drug Dictionary). These TCRs are typically induced by multi-peptide vaccines or utilized in TCR-engineered T-cell (TCR-T) therapies restricted to the HLA-A*2402 allele, a common MHC class I molecule in Asian populations (NCT00561275, MDPI: 10.3390/vaccines10010001). By recognizing these specific epitopes, the TCRs mediate the activation of cytotoxic T-lymphocytes (CTLs) that selectively destroy both the tumor cells and the supporting blood vessels, offering a synergistic therapeutic approach to inhibit tumor growth and progression.
