The CD3δε (CD3 delta-epsilon) heterodimer is a crucial component of the T-cell receptor (TCR) complex, playing a fundamental role in signal transduction following antigen recognition (UniProt P04234, P07766). It consists of two invariant polypeptide chains, delta (δ) and epsilon (ε), which associate with the TCR alpha/beta or gamma/delta chains to facilitate T-cell activation and development (NCBI Gene 915, 916). In therapeutic contexts, CD3δε is a primary target for bispecific antibodies (e.g., BiTEs) designed to redirect T-cells to eliminate malignant cells by cross-linking the TCR with tumor-associated antigens (PubMed: 32661138). While highly effective in treating various hematologic malignancies, targeting this complex can lead to systemic immune activation, resulting in safety concerns such as cytokine release syndrome (CRS) and neurotoxicity (StatPearls: NBK545235). Additionally, monoclonal antibodies against CD3 have been utilized for immunosuppression in organ transplantation and autoimmune disorders by modulating or depleting T-cell populations (DrugBank: DB00075).
