Overview

The T-cell surface glycoprotein CD3 gamma chain (CD3G) is an essential transmembrane subunit of the T-cell receptor (TCR)-CD3 complex, primarily expressed on the surface of mature T lymphocytes and thymocytes (UniProt P09693; Wikipedia). It plays a pivotal role in the adaptive immune response by coupling extracellular antigen recognition to intracellular signaling pathways through its cytoplasmic immunoreceptor tyrosine-based activation motif (ITAM) (NCBI Gene 917; UniProt P09693). Beyond signal transduction, CD3G is critical for the assembly, trafficking, and stable surface expression of the TCR complex, utilizing a di-leucine-based motif to regulate receptor internalization and recycling (PubMed 8187769; Open Targets). Mutations in the CD3G gene lead to T-cell immunodeficiency (IMD17), characterized by impaired T-cell development and increased susceptibility to infections and autoimmunity (NCBI Gene 917; PubMed 25920998). While most established CD3-targeted therapies, such as Muromonab-CD3 and various bispecific antibodies, primarily engage the CD3 epsilon subunit, CD3G is increasingly recognized as a distinct therapeutic target for fine-tuning T-cell activation in autoimmune diseases and oncology (Cusabio; Creative Biogene). Its expression levels and genetic variants also serve as valuable biomarkers for monitoring immune status and predicting treatment outcomes in sepsis and cancer (PubMed 25920998; NIH).

Mechanism of action

CD3G acts as a signal transduction subunit within the TCR-CD3 complex. Upon antigen binding to the TCR, the immunoreceptor tyrosine-based activation motifs (ITAMs) in the cytoplasmic tail of CD3G are phosphorylated by Src-family kinases like Lck and Fyn (UniProt P09693; PubMed 2470098). This phosphorylation recruits ZAP-70, initiating downstream signaling cascades including the Ras-MAPK, PKC, and calcium-NFAT pathways, leading to T-cell activation and proliferation (Creative Biogene; PubMed 25920998). Additionally, CD3G regulates TCR expression levels through a di-leucine-based sorting motif that mediates constitutive receptor internalization and recycling (UniProt P09693; PubMed 8187769).
