Overview

T-lymphocyte activation antigen CD80 mRNA is the genetic template for the CD80 protein, a critical costimulatory ligand also known as B7-1 (NCBI Gene: 941) [1]. Expressed primarily on activated B cells and other antigen-presenting cells, the CD80 protein provides essential signals for T-cell activation and survival by binding to the CD28 receptor [2]. It also interacts with the inhibitory receptor CTLA-4 to attenuate immune responses, maintaining immunological self-tolerance [3]. In therapeutic development, CD80 mRNA is targeted by RNA interference (siRNA) or antisense technologies to suppress T-cell-mediated inflammation in autoimmune diseases and transplant rejection [4]. Conversely, synthetic CD80 mRNA is being explored in cancer immunotherapy to induce the expression of costimulatory molecules on tumor cells, thereby enhancing T-cell-mediated anti-tumor activity [5]. While current clinical therapies like Abatacept and Belatacept target the CD80 protein directly, mRNA-based approaches offer a novel mechanism for modulating the immune system at the pre-translational level [6].
