Tax1-binding protein 3 (TAX1BP3), widely known as TIP-1, is a scaffold protein distinguished by its single PDZ domain that facilitates critical protein-protein interactions (UniProt: Q9ULV0). It functions as a key regulator in multiple signaling pathways, most notably the Wnt/beta-catenin and RhoA pathways, which are essential for maintaining cell polarity and migration (PubMed: 22431518). In the context of oncology, TIP-1 is frequently upregulated in various cancers, including glioblastoma and non-small cell lung cancer, where it contributes to tumor progression and metastasis (PubMed: 25665555). A unique feature of TIP-1 is its translocation to the cell surface following ionizing radiation, providing a specific target for radiotherapy-guided drug delivery (PubMed: 18413744). Current drug development efforts focus on small molecules and peptides designed to bind the PDZ pocket, thereby blocking the recruitment of downstream effectors and inhibiting tumor growth (PubMed: 21810415). Although still in the preclinical stage, targeting the TIP-1 PDZ domain represents a novel approach to precision oncology and molecular imaging.
