Overview

TBX2 antisense RNA 1 (TBX2-AS1) is a long non-coding RNA (lncRNA) located on chromosome 17q23.2, transcribed from the antisense strand of the TBX2 gene (Stanford University, 2022; ResearchGate, 2025). It primarily functions as a cis-regulator of the TBX2 transcription factor, which is essential for embryonic development and hair cell differentiation in the inner ear (Journal of Medical Genetics, 2025). In the context of oncology, TBX2-AS1 is frequently overexpressed and acts as an oncogene in various cancers, such as osteosarcoma and hepatocellular carcinoma, where it promotes tumor progression, metastasis, and resistance to therapy (NIH, 2022). Research indicates that knockdown of TBX2-AS1 leads to a significant reduction in TBX2 expression, thereby inhibiting the oncogenic pathways driven by this transcription factor (ResearchGate, 2025). Additionally, disruptions in the TBX2-AS1 locus have been associated with sensorineural hearing loss and other rare germline disorders (Journal of Medical Genetics, 2025). While no small-molecule drugs currently target TBX2-AS1 directly, it is considered a promising candidate for nucleic acid-based therapies, such as antisense oligonucleotides (ASOs) or siRNAs (NIH, 2022).
