TEAD1-4 are a family of highly conserved **transcription factors** characterized by a TEA (TEAD) DNA-binding domain and a coactivator-binding domain. They act as the main terminal effectors in the Hippo signaling pathway, orchestrating key cellular events such as cell proliferation, differentiation, organ size regulation, and stem cell biology in both physiological and pathological settings. TEAD proteins exert their function by binding DNA and recruiting transcriptional co-activators—including YAP and TAZ—to control the expression of genes that drive cell growth and survival. Abnormal TEAD activity is closely associated with tumorigenesis, including EMT, metastasis, and chemoresistance, making these proteins promising therapeutic targets under active investigation.
