Overview

Testis expressed 14, intercellular bridge forming factor (TEX14) is a scaffolding protein critically required for the formation and maintenance of intercellular bridges between germ cells during spermatogenesis. It localizes to intercellular bridges formed after incomplete cytokinesis, ensuring cytoplasmic communication and synchronization among developing sperm. TEX14 loss-of-function mutations cause male-specific infertility due to the absence of these bridges, leading to arrest of spermatogenesis before meiotic division. TEX14 is not a protein kinase, despite bearing a kinase-like domain, and is instead believed to serve as a scaffold in bridge assembly. There are no drugs that target TEX14, and it is not considered a conventional therapeutic target, though it may serve as a biomarker in certain settings, such as male infertility or as a cancer/testis antigen in oncology.
