Overview

TANC1 is a large scaffold protein prominently expressed in the adult brain, particularly in postsynaptic densities where it organizes synaptic complexes, influencing dendritic spine maintenance and the strength/plasticity of excitatory synapses[1][2][5]. It contains tetratricopeptide repeat, ankyrin repeat, and coiled-coil domains, facilitating protein-protein interactions and possibly regulating key neuronal signaling pathways, including those of the glutamate receptor, planar cell polarity, and Hippo signaling[1][2]. Functional and genetic studies suggest TANC1 is important in neurodevelopment and synaptic structure, with mutations and disruption implicated in intellectual disability and autism spectrum disorder[1][2][4]. While TANC1 is not a direct drug target, understanding its scaffolding role in neuronal architecture is valuable for elucidating the molecular basis of several neurodevelopmental conditions[1][2][5].
