Overview

TGF-beta activated kinase 1 (MAP3K7) binding protein 3 (TAB3) is an adaptor protein that is essential for the activation of JNK and NF-kappa-B signaling pathways. This protein specifically recognizes Lys-63-linked polyubiquitin chains, organizing large signaling complexes that promote autophosphorylation of MAP3K7/TAK1 and subsequent activation of IKK and related cascades. TAB3 plays a crucial role in mediating signal transduction in response to pro-inflammatory cytokines such as TNF and IL-1, functioning in innate immunity, inflammatory responses, and cellular stress signaling. Dysfunction or altered expression of TAB3 has been implicated in several pathological states, including inflammatory diseases, cancers, and certain genetic skeletal disorders[1][3][5].
