Overview

Thymidylate synthase (TS) is a crucial enzyme in the de novo synthesis of dTMP, a building block for DNA. In fungi, such as Candida albicans, TS is essential for DNA replication and repair. It catalyzes the reductive methylation of dUMP to dTMP, using 5,10-methylenetetrahydrofolate as a cofactor. As TS is essential for fungal survival and proliferation, it has become a promising antifungal target. Inhibitors of fungal TS can lead to "thymineless death".
