TLE family member 3, transcriptional corepressor (TLE3), is a member of the Groucho/transducin-like enhancer (Gro/TLE) family of transcriptional corepressors that does not itself bind DNA but modulates gene expression by interacting with a variety of transcription factors[1][6][7]. TLE3 participates in several developmental and metabolic signaling pathways including Wnt, Notch, and PPARγ, acting as a key regulator of adipogenesis and memory B cell differentiation[2][4][5]. In adipogenesis, TLE3 integrates PPARγ and Wnt signaling, enhancing PPARγ-mediated gene activation while suppressing anti-adipogenic Wnt signals, which can influence systemic insulin sensitivity[2]. TLE3 also interacts with transcription factors such as Hhex to promote memory B cell development[4]. In cancer, particularly lung cancer, higher TLE3 expression has been linked to improved patient outcomes[1]. There are currently no approved drugs that directly target TLE3, and its manipulation for therapeutic purposes poses potential challenges due to its broad effects on transcriptional regulation.
