Overview

TNF receptor-associated factor 2 (TRAF2) is a multi-functional adapter protein and E3 ubiquitin ligase that plays a central role in the signal transduction of the tumor necrosis factor receptor (TNFR) superfamily (UniProt P39429). It is essential for the activation of the canonical NF-kappaB and JNK signaling pathways, which govern cell survival, inflammatory responses, and stress-induced apoptosis (PubMed: 23604121). In various malignancies, TRAF2 is frequently overexpressed, where it promotes oncogenesis by inhibiting pro-apoptotic signaling and enhancing tumor cell proliferation (PubMed: 28234355). While direct inhibitors of TRAF2 are still in early development, it is a primary component of the cIAP-TRAF complex targeted by SMAC mimetics like Birinapant and Xevinapant, which induce the degradation of these complexes to promote cancer cell death (PubMed: 24658274). Beyond oncology, TRAF2 is implicated in the pathogenesis of chronic inflammatory and autoimmune diseases due to its role in cytokine-mediated signaling (PubMed: 30108334). Consequently, TRAF2 represents a significant therapeutic node for modulating immune and apoptotic pathways in diverse disease contexts.
