Overview

TNF receptor-associated factor 4 (TRAF4) is a member of the TRAF family of scaffold proteins that also possesses E3 ubiquitin ligase activity (UniProt Q9BUZ4). Unlike other TRAF proteins that primarily mediate immune and inflammatory signaling, TRAF4 is uniquely involved in regulating cell-cell adhesion, tight junctions, and the epithelial-mesenchymal transition (EMT) (PubMed: 23934152). It is frequently upregulated in various human cancers, including breast, lung, and prostate carcinomas, where it acts as an oncogene by potentiating signaling pathways such as TGF-beta, Wnt/beta-catenin, and Hippo (PubMed: 28234355). Targeting TRAF4 mRNA via RNA interference (siRNA) or antisense oligonucleotides (ASOs) represents a promising therapeutic strategy to suppress tumor growth and metastasis (PubMed: 30104359). Current research focuses on overcoming the challenges of delivering these RNA-based agents to tumor sites while minimizing off-target effects. While no TRAF4-targeted therapies are currently FDA-approved, its role as a key nodal point in oncogenic signaling makes it an attractive candidate for precision medicine.
