Overview

The Toll-like receptor 4 (TLR4) mRNA 3' untranslated region (UTR) is a critical regulatory segment of the TLR4 transcript that governs the stability and translation of the receptor protein. TLR4 is a central component of the innate immune system, primarily responsible for recognizing lipopolysaccharides (LPS) from Gram-negative bacteria and initiating inflammatory signaling pathways (Taganov et al., PNAS 2006). The 3' UTR contains multiple binding sites for microRNAs (miRNAs), such as miR-146a and miR-223, and RNA-binding proteins that act as post-transcriptional rheostats to prevent excessive inflammation (He et al., 2014). Dysregulation of these regulatory interactions is linked to chronic inflammatory conditions, sepsis, and neuropathic pain, where over-expression of TLR4 drives pathological cytokine storms. Therapeutic strategies targeting this region, such as miRNA mimics or antisense oligonucleotides, aim to downregulate TLR4 expression to mitigate pathological inflammation. Consequently, the TLR4 mRNA 3' UTR represents a high-value target for precision modulation of the immune response in various inflammatory and autoimmune contexts.
