Overview

Topless-related protein 2 (RELK2) is a member of the TOPLESS (TPL) family of transcriptional corepressors in maize (Zea mays). It plays a critical role in plant development by mediating the repression of target genes involved in hormone signaling and meristem identity. RELK2 functions by interacting with various transcription factors, such as RAMOSA1, through their EAR (Ethylene-responsive element binding factor-associated Amphiphilic Repression) motifs, subsequently recruiting histone deacetylases to modify chromatin structure and silence gene expression (Gallavotti et al., 2010). This corepressor is essential for maintaining proper inflorescence architecture and regulating the transition between different types of meristems. While not a traditional human therapeutic target, RELK2 is a significant target for agricultural biotechnology aimed at optimizing crop yield and plant morphology. Understanding its interaction with plant hormones like auxin and jasmonate provides insights into how plant growth can be chemically or genetically manipulated to improve agricultural productivity (Liu et al., 2019; Espinosa-Ruiz et al., 2017).

Mechanism of action

RELK2 acts as a transcriptional corepressor by interacting with EAR-motif containing transcription factors to recruit histone deacetylases, thereby inducing chromatin remodeling and gene silencing (Gallavotti et al., 2010; Liu et al., 2019).
