Overview

The TP53 R175H peptide presented by HLA-A*02:01 is a tumor-specific neoantigen complex that forms when a peptide containing the hotspot cancer-associated R175H missense mutation of the p53 protein (sequence: HMTEVVRHC) is loaded into the peptide-binding groove of the MHC class I molecule HLA-A*02:01 on the surface of tumor cells. This complex can be specifically recognized by engineered T cell receptors or TCR mimic antibodies, enabling the immune system or immune therapies to selectively target tumor cells harboring this mutation, while sparing normal cells. The complex is structurally unique due to the anchoring and presentation of the mutant His175 residue, which is exposed and can be targeted for immunotherapy. Clinical interest focuses on its use for personalized cancer vaccines and adoptive cell therapies, with the safety and efficacy determined by the specificity of immune recognition for this neoantigen.
