Trace amine-associated receptor 5 (TAAR5) is a G protein-coupled receptor (GPCR) that serves as a high-affinity sensor for diamines, most notably cadaverine and trimethylamine. While primarily recognized for its role in the olfactory system—where it mediates the perception of decaying organic matter and triggers innate aversive behaviors—recent evidence highlights its expression in the central nervous system, particularly in limbic regions such as the amygdala and hippocampus. In these areas, TAAR5 is involved in modulating emotional states, social behaviors, and adult neurogenesis. Dysregulation of TAAR5 signaling has been linked to psychiatric conditions, including depression and anxiety, as well as neurodegenerative diseases like Parkinson's. Therapeutic interest in TAAR5 is growing, with research focusing on the development of antagonists as potential treatments for mood disorders. Cadaverine, the primary endogenous ligand, is a product of lysine decarboxylation and serves as a significant biomarker for bacterial overgrowth and certain malignancies, such as breast and lung cancer.
