Transcription activator BRG1 (SMARCA4) is the central catalytic ATPase subunit of the SWI/SNF family of chromatin-remodeling complexes. Using ATP-derived energy, BRG1 disrupts chromatin architecture at target promoters to regulate gene expression by controlling DNA accessibility. The protein forms part of multiple multi-protein complexes that regulate transcription, DNA replication, DNA repair, and cellular differentiation through chromatin remodeling. BRG1 functions as both a transcriptional activator and repressor, often working in conjunction with nuclear receptors, histone-modifying enzymes, and transcriptional coregulators to control gene activity in various physiological contexts. BRG1 is the most frequently mutated chromatin remodeling enzyme in human cancers, with mutations identified in lung, adrenal gland, pancreatic, and medulloblastoma cancers, among others. These cancer-associated mutations preferentially target the ATPase domain and act in a dominant manner to alter chromatin regulatory function, enabling cancer cells to evade differentiation-inducing therapies. Beyond cancer, BRG1 is critical for normal development; loss of functional BRG1 prevents embryonic implantation and causes infertility through disruption of spermatogenesis. Emerging drug development efforts include BRG1/BRM inhibitors such as FHD-286, positioning BRG1 as an emerging therapeutic target for cancer treatment.
