Overview

JUN and FOS are components of the Activator Protein-1 (AP-1) transcription factor complex, which converts extracellular signals into changes in gene expression related to proliferation and apoptosis (UniProt P05412, P01100). MAPK1, also known as ERK2, is a serine/threonine kinase that serves as a critical node in the MAPK/ERK pathway, phosphorylating numerous targets including JUN and FOS to promote cell cycle progression (UniProt P28482). Together, these molecules form a highly interconnected signaling network frequently dysregulated in human cancers and chronic inflammatory diseases. Because this target string represents multiple distinct proteins rather than a single therapeutic target or a single stable biochemical complex, it is categorized as a multi-target signaling set.

Mechanism of action

Inhibition of mitogen-activated protein kinase activity (MAPK1), inhibition of proteasomal degradation of IkappaB to prevent RELA nuclear translocation, and disruption of AP-1 (JUN/FOS) transcriptional activity.
