Overview

Transcription initiation factor TFIID subunit 1-like (TAF1L) bromodomain 2 is the second of two tandem acetyl-lysine binding modules located within the TAF1L protein, a testis-specific paralog of the general transcription factor TAF1 [1]. As a component of the TFIID complex, TAF1L-BD2 plays a critical role in recognizing specific histone acetylation marks, which facilitates the recruitment of the transcriptional machinery to gene promoters [2]. While TAF1L is primarily known for its essential role in male germ cell development and spermatogenesis, its aberrant expression has been implicated in various malignancies, including prostate cancer and certain leukemias [3, 4]. Small molecule inhibitors targeting the bromodomains of TAF1L, such as GNE-049, aim to disrupt these chromatin interactions to suppress the expression of oncogenic drivers [5]. However, the high degree of sequence identity between the bromodomains of TAF1L and TAF1 poses a significant challenge for the development of selective inhibitors [2]. Consequently, therapeutic intervention may lead to safety concerns regarding reproductive health and potential systemic toxicity from the disruption of essential TAF1-mediated transcription in non-target tissues [6].

Mechanism of action

Competitive inhibition of acetylated lysine binding to the bromodomain pocket, which prevents the recruitment of the TFIID complex to chromatin and subsequently inhibits gene transcription.
