Overview

Transducer of ERBB2, 2 (TOB2) is an anti-proliferative protein in humans, encoded by the TOB2 gene. It is a member of the BTG/Tob family of antiproliferative proteins that negatively regulate cell cycle progression, mainly by inhibiting the transition from G0/G1 to S phase[1][2][3][4]. TOB2 acts as a tumor suppressor by restraining cell growth, partly through regulating mRNA turnover and promoting global mRNA decay via its interaction with the CAF1-CCR4 deadenylase complex and the poly(A)-binding protein (PABP)[2]. Its function and stability are further modulated by phosphorylation and ubiquitin–proteasome system–mediated degradation[2]. TOB2 protein is regulated by various oncogenic microRNAs (e.g., miR-378, miR-362-3p), which decrease its expression to enhance cellular proliferation in cancers such as hepatocellular carcinoma[2]. Altered expression or regulation of TOB2 is implicated in pathological cell proliferation and may play a broader role in tumorigenesis, but it is not currently considered a direct therapeutic drug target and has no known interacting drugs or established biomarker utility[3].
