Target intelligence / Profile preview
Transforming growth factor beta 1 (TGFB1) mRNA is the messenger RNA transcript that encodes the TGF-beta 1 protein, a potent pleiotropic cytokine (Source: UniProt P01137). TGF-beta 1 is a master regulator of the immune system, cell proliferation, and the production of the extracellular matrix, making its mRNA a critical point of therapeutic intervention (Source: PMC6711358). In pathological states, overproduction of TGF-beta 1 driven by high mRNA levels leads to extensive tissue fibrosis and promotes tumor immune evasion and metastasis (Source: PMC4012445). Targeting the mRNA directly via antisense oligonucleotides (ASOs) or small interfering RNAs (siRNAs) provides a method to specifically downregulate TGF-beta 1 expression, bypassing some of the complexities of receptor-level inhibition (Source: PMC4423186). Therapeutic candidates like ISTH1047 and STP705 have been developed to treat aggressive tumors and fibrotic conditions by silencing the production of this growth factor at the source (Source: ResearchGate DOI:10.1158/1078-0432.CCR-17-3024). This approach is particularly valuable in oncology and fibrotic diseases where systemic protein inhibition might lead to adverse effects.
