Overview

The Transient Receptor Potential (TRP) channel family is a large group of non-selective cation channels that function as versatile cellular sensors for a wide range of physical and chemical stimuli (guidetopharmacology.org). In mammals, the family consists of 28 members divided into six subfamilies: TRPC, TRPV, TRPM, TRPA, TRPP, and TRPML (nih.gov). These channels are critical for sensory perception, including the detection of temperature, pain, taste, and mechanical stress, as well as for maintaining ion homeostasis and regulating cellular signaling pathways (frontiersin.org). Due to their widespread expression and involvement in various physiological processes, TRP channels are implicated in numerous diseases such as chronic pain, inflammation, cancer, and cardiovascular disorders (nih.gov). Consequently, they have become major targets for drug discovery, with several agonists and antagonists currently in clinical development for conditions like neuropathic pain and respiratory diseases (guidetopharmacology.org). However, therapeutic targeting faces challenges such as maintaining selectivity and managing side effects like altered body temperature regulation (nih.gov).
