Overview

Transmembrane protease, serine 11F (TMPRSS11F) is a type II transmembrane serine protease mainly found at the cell surface of diverse epithelial tissues and exocrine glands[1][2][3][5]. It plays an essential role in epidermal barrier formation and maintenance, facilitating proteolysis required for normal skin function and hydration[1][2][3]. Studies in knockout mice show its importance in preventing trans-epidermal fluid loss, especially in neonates[1][2]. Abnormal expression in cancer, specifically acute myeloid leukemia, is correlated with poor prognosis and promotes cancer cell invasion via activation of matrix metalloproteinase-2[1]. There are no documented drug interactions or approved targeted therapies, but the molecule is being explored as a cell surface biomarker and as a candidate therapeutic target in oncology[1][2][3][5].

Mechanism of action

not established for approved drugs; mechanism described in cancer is activation of matrix metalloproteinase-2 facilitating cancer invasion[1]

03

Biological functions

Proteolysis (serine-type endopeptidase activity)Establishment and maintenance of skin/epidermal barrierHomeostasis (body fluid balance)Cell surface marker (in AML)
04

Disease associations

Cancer (acute myeloid leukemia; promotes cell invasion/tumor growth when aberrantly expressed)Skin diseases (e.g., autosomal recessive congenital ichthyosis)
05

Safety considerations

no notable direct safety concerns; knockout studies in mice show no gross abnormalities except under dehydration or stress conditions[2]therapeutic targeting in cancer may involve off-target effects due to protease activity
06

Biomarkers

aberrant cell-surface expression in AML may serve as a diagnostic/prognostic cell surface marker
