Tribbles homolog 2 (TRIB2) is a member of the Tribbles family of pseudokinases, characterized by a conserved kinase-like domain that lacks essential catalytic residues for phosphate transfer (UniProt P56746). Despite its lack of catalytic activity, TRIB2 functions as a critical scaffold protein and an adapter for E3 ubiquitin ligases, such as COP1, facilitating the degradation of key transcription factors like C/EBPα (PubMed: 25201210). In oncology, TRIB2 is recognized as a potent oncogene, particularly in acute myeloid leukemia (AML), where its overexpression promotes cell survival and blocks myeloid differentiation (PubMed: 17632521). It also plays significant roles in solid tumors, including melanoma and liver cancer, by modulating the MAPK and AKT signaling pathways (PubMed: 23603136). Therapeutic strategies targeting TRIB2 focus on small molecule inhibitors that bind its pseudokinase domain to either destabilize the protein or prevent its interaction with downstream effectors (PubMed: 30104376). Additionally, TRIB2 has been identified as an autoantigen in narcolepsy, highlighting its diverse biological relevance beyond cancer (PubMed: 20107029).
