Overview

TRIM52 antisense RNA 1 (head to head) (abbreviated as TRIM52-AS1) is a primate/human-specific long non-coding RNA (lncRNA) gene located on chromosome 5. It is transcribed as an antisense RNA opposite the TRIM52 protein-coding locus. TRIM52-AS1 functions as an epigenetic regulator, mainly by binding and sequestering microRNAs such as miR-361-5p and miR-577, thereby modulating the expression of target genes associated with cell proliferation, apoptosis, and radioresistance. TRIM52-AS1 is implicated in the progression of multiple cancers (including colorectal, hepatocellular, renal cell carcinoma, and diffuse large B-cell lymphoma), where its upregulation can promote tumor growth, reduce apoptosis, and worsen prognosis. Experimental inhibition or knockdown of TRIM52-AS1 increases cancer cell sensitivity to radiation and suppresses tumor cell viability, indicating therapeutic potential. No approved drugs currently target TRIM52-AS1, but it may serve as a candidate biomarker for cancer diagnosis, prognosis, and therapeutic response.

Mechanism of action

TRIM52-AS1 acts as a miRNA sponge for miR-361-5p and miR-577, modulating downstream gene expression (e.g., TRIM52, IGFBP3). In experimental settings, RNA interference or antisense oligonucleotides may alter TRIM52-AS1 function.
